Is it grammatically correct to say, “what I understood the most out of the book of Genesis was...”

    Yes. This is adequate as far as it goes, although it could be stated differently.

    It's  correct lacking naturalness required for a sentence.

    Yes, it is grammatically correct, but it is incredibly awkward.
